The ride, part of Swindon & Wiltshire Pride's celebration weekend, will raise money for next year's festival.
This year's was cancelled due to the covid-19 pandemic.
Pride chair Jo Sharpe said: "Swindon & Wiltshire Pride, like many charities, will struggle in the coming months and years due to the pandemic.
"As a committee, we decided very early on to divert our fundraising efforts to the Swindon Food Collective during lockdown.
"The 8 August would have been Swindon & Wiltshire Pride 2020. The committee are still keen to have a presence within the community, so instead of the Pride Festival the committee are going to do a bike ride which gives us the chance to spread the rainbow around Swindon, and also to thank our sponsors and the community for their support.
"So if you see us on our ride, please wave, cheer, say hello or even blow your horn!"
The Ride for Pride will start at The Waiting Room pub in Fleet Street at 10.30am.
The riders will make their way to Rainbow Walk near the Wyvern Theatre and Swindon Borough Councils Euclid Street headquarters before heading for Old Town.
The route will take in the Town Gardens, which is the home of Pride, followed by Intel, Nationwide Building Society - the main sponsors for 2021 - and Thames Water before heading back to the starting point.
